功能与背景

This gene encodes a cyclin-dependent kinase inhibitor, which shares a limited similarity with CDK inhibitor CDKN1A/p21. The encoded protein binds to and prevents the activation of cyclin E-CDK2 or cyclin D-CDK4 complexes, and thus controls the cell cycle progression at G1. The degradation of this protein, which is triggered by its CDK dependent phosphorylation and subsequent ubiquitination by SCF complexes, is required for the cellular transition from quiescence to the proliferative state. Mutations in this gene are associated with multiple endocrine neoplasia type IV (MEN4). [provided by RefSeq, Apr 2014]

Wild-type (WT) and CDKN1B knockout (KO) HeLa cell extracts (30 μg) were separated by 12% SDS-PAGE, and the membrane was blotted with p27 Kip1 antibody [HL2508] (GTX638869) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.

Various whole cell extracts (30 μg) were separated by 12% SDS-PAGE, and the membrane was blotted with p27 Kip1 antibody [HL2508] (GTX638869) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.

The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody. p27 Kip1 antibody [HL2508] detects p27 Kip1 protein at cytoplasm and nucleus by immunofluorescent analysis.
Sample: MCF-7 cells were fixed in 4% paraformaldehyde at RT for 15 min.
Green: p27 Kip1 stained by p27 Kip1 antibody [HL2508] (GTX638869) diluted at 1:500.
Red: alpha Tubulin, a cytoskeleton marker, stained by alpha Tubulin antibody [GT114] (GTX628802) diluted at 1:1000.
Blue: Fluoroshield with DAPI (GTX30920).
