Regulator of G-protein signaling 4 (RGS4) belongs to the structurally smallest (i.e., B/R4) family of G protein GTPase-activating proteins (GAPs) in the RGS superfamily. RGS proteins modulate a plethora of G protein-coupled receptor (GPCR)-triggered signaling cascades via their GAP function on G protein α subunits, essentially increasing their GTPase activity to convert Gα proteins into their inactive GDP-bound form. RGS4 is expressed primarily in the nervous system and heart, and is linked to several neuronal GPCR pathways (1). Recent studies have also revealed RGS4 to be a potential therapeutic target for both human heart failure (HF) and atrial fibrillation (AFib) (2).
